Defense
When you see a gold line come up in the Chat with your character's name in it....
The Table: Deqqie goes to use Charcoal Grip, 13 against Cochise (Range:23" Speed:6).
It's probably best to start thinking about defending against the attack, aka “setting a defense.” The easiest way is to simply punch a number into the box on the far left of the defensive line and then clicking (which is normally silver but lights up blue when it has a decent number to work with).

Of course, with defense you will never simply pull a number out of thin air. Instead you need a value from your character sheet, which means clicking the silver button to the right of the defensive value box and summoning this pop-up....
It's use is pretty straight forward. Click on a link the pop-up will close and set your defense - just as if you had pumped a number into the Defensive line and clicked (since there are no modifiers to add it can safely do this). In the Game Chat will appear a line like this....
The Table: Cochise sets a Total Defense of 89.
And in everyones table controls will appear a reiteration of this just so we don't forget what the current defense is set at.

If anyone sets a new defense, this one will be replaced by it.
A few last tips about the Defensive pop-up. In combat you will almost always use the Quick Armor links at the top of the page, but if you need to calculate your armor while missing a few pieces (like a helmet) clicking and unclicking the checkboxes in the Armor Table will change the total score at the bottom. Clicking on that link will return the total score.
Generic Defenses are primarily used by the Director to oppose Ability and Skill checks and such. They are shown to the players as a guide to what constitutes a strong or a weak defense in a completely average situation. Of course, there is nothing quite so uncommon as a completely average situation in gaming.
